Niğde ilinde yetiştirilen fasulye (Phaseolus vulgarıs L.) bitkilerinde potyvirus ve fitoplazma hastalıklarının araştırılması

Sixty potyvirus-positive samples were selected to be tested by RT-PCR. Fifty four samples were found to be infected with Bean common mosaic virus (BCMV), one sample infected with Bean yellow mosaic virus (BYMV). The sequenced isolate 186 showed 89% identity to a BYMV isolate from pea in Germany deposited in NCBI. In this study, ten common bean cultivars were inoculated with a BCMV isolate selected based on its high level of virulence trait, and their symptom appearance was evaluated after inoculation. Based on the phenotypic evaluations and PTA-ELISA screening, two cultivars were found to be susceptible to BCMV, while the eight cultivars were discovered to be resistant. Also, SCAR (SW-13, SBD-5, and ROC11) and CAPS (eIFE4) markers were used to detect resistance genes in 22 common bean cultivars. Results showed that 11 involved the I gene-specific product with SW-13 marker, while 13 contained the recessive bc-12 gene-specific product with SBD-5 marker. On the other hand, the recessive bc-3 gene was found with ROC11 marker but not with eIFE4 in common bean cultivars tested. Moreover, 56 symptomatic common bean samples were tested for phytoplasma detection, no phytoplasma was detected in the samples tested. To our knowledge, this is the first study in the attempt at molecular identification of phytoplasma diseases in common beans in Türkiye.
