Abstract (EN)
Nigella sativa, a plant known as nigella. In general this type of plant used as a spice. This, in the Mediterranean region and India, including Pakistan and Afghanistan is a spice that grows in the West Asian countries. Nigella sativa seeds, today, asthma and other lung diseases, gastrointestinal disorders (eg; indigestion, diarrhea), and is used in the treatment of hyperlipidemia. Also known for antioxidant effects and immune regulatory activity.Carbonic anhydrase (E.C. 4.2.1.1.), the center of a metalloenzyme with Zn+2.CO2 in vertebrates, is a substrate for this enzyme. Cells and between cells is an enzyme that catalyzes the transformation of liquid CO2 and H2CO3.In this study, Nigella sativa, was investigated inhibitory effects on the enzyme carbonic anhydrase. For that purpose, human erythrocyte carbonic anhydrase isozymes (HCA-I and HCA-II) were purified separately by affinity chromatography. Later, Nigella sativa (black seed) homogenates prepared. Homogenates of human erythrocyte CA enzymes with different concentrations on the inhibitory effect of Nigella sativa were investigated. Hydratase and esterase activities of the studies utilized for determining the activity of carbonic anhydrase. Nigella sativa certain concentrations (0.25% to 0.5%, 0.1%, 0.2%, 0.3%) of human erythrocytes was determined that the inhibitory effect of CA hydratase and esterase activity. % Activity-[I] for the inhibition effect homogenates IC50 values were plotted graphs. HCA-I shows the inhibition of esterase activity of all concentrations IC50 values (concentration of inhibitor that reduces by half the duration of enzymatic reaction), 0,2101.104 ? M-0,0492.104 ? M was found between. 0.1%, 0.2%, 0.3% HCA-I concentrations on the inhibitory effect of Nigella sativa homogenates Ki values were examined; 4,290.102 ? M-3,980.102 ? M are among the.
