Abstract (EN)
Norfloxacin (NFX), the broad-spectrum antimicrobial-acting analogue of nalidixic acid, is a fluoroquinolone that has strong activity against gram-positive and gram-negative bacteria, while having limited activity against anaerobes. Topical ocular drug application is one of the most preferred ways to treat anterior segment diseases of the eye due to its rapid effect and high patient compliance. The bioavailability of the active substance in the eye is low due to the anatomical and physiological characteristics of the eye. Within the scope of our thesis studies, nanoparticles were prepared using polylactic-co-glycolic acid (PLGA), Chitosan and Eudragit L-30 D-55 polymers by NFX nanocomposition method in order to develop a formulation of NFX nanoparticles for ocular application. It was aimed to prolong the duration of the active substance with nanoparticular systems and to provide an effective treatment with a lower dose of the active substance. The physicochemical characteristics of the prepared formulations were examined in detail using in vitro analysis methods. When the results of the analyzes were evaluated, it was determined that the formulations successfully provided extended release of the active substance at ocular pH. As a result of the antifungal activity studies, it was concluded that an effective treatment can be provided at lower doses.
