Abstract (EN)
Ameloblastomas and odontogenic keratocysts share many clinical features in common such as aggressiveness, high recurrence rates and radical management options. Understanding the pathogenesis and biological aspects of these tumours would improve the success of diagnose and treatment procedures.It is necessary for these tumours to have a ?lytic? ability in their aggressive growth potential. Tissue degrading factors include cytokines such as IL-1? and IL-6 which are the most effective ones in bone remodelling.The aim of this study was to exhibit the reasons of their high recurrence rates and growth potentials of ameloblastomas and keratocystic odontogenic tumours by investigation of the expression and gene polymorphism of IL-1? and IL-6.This study included formaline fixed parafine embedded 25 ameloblastomas, 41 parakeratinized keratocysts (keratocystic odontogenic tumour) and 8 orthokeratinized keratocysts cases archived in Gazi University Faculty of Dentistry Oral Pathology Department. All histological slides were stained immunohistochemically to show the expression of IL-1? and IL-6. Restriction fragment length analysis was used to investigate the cytokine gene polymorphism.The higher expression rates of IL-1? and IL-6 were associated with tumour size in ameloblastomas and with cyst wall thickness in keratocysts. Our results suggest that polymorphism of IL-1? -889 region is not a risk factor in the development of these tumours. But it was showed that IL-1? (-889) T polymorphism increases the IL-1? expression. According to the allele frequencies T allele was higher in ameloblastomas. Due to the troubles in amplification IL-6 -174 region, polymorphism could not be analysed.
