Investigation of possible therapeutic and preventive effect of omentin on absance epilepsy

Epilepsy is one of the frequently observed central nervous system disorder. Ethosuximide (ETX) is a drug of choice for the symptomatic treatment of absence seizures. Omentin is an adipocytokine that is abundantly expressed in visseral fat tissue. Omentin increases AMP-activated protein kinase (AMPK) phosphorylation, GABAB receptors are intimately associated with 5'AMP- dependent protein kinase (AMPK). GABAB receptors are heterodimeric G protein-coupled receptors composed of R1 and R2 subunits. GABAB receptors mediate slow prolonged inhibition in the brain by activating postsynaptic inwardly rectifying K+ channels (GIRKs) and inactivating presynaptic voltage-goted Ca++ channels. For this experiment male, having weight of 250- 350 gr WAG/ Rij (n= 16) and Wistar Albino (n=40) rats were used. After stereotaxic surgical procedures, all rats were returned ready for direct measurement of cortical EEG. Subject were seperated into seven groups. Group A1: Control, Group A2: Pentylenetetrazol (PTZ), Group A3: Ethosuximide + PTZ, Group A4: Ethosuximide + PTZ combined with Omentin, Group A5: Omentin+ PTZ, Group A6: PTZ+ Omentin as a result of the implementation of six groups (n=8) in Wistar Albino rats. Ethosuximide (125mg/kg/3 day), Omentin (8 μg/ kg/3 day) and PTZ (40 mg/kg) administered intraperitoneally. DDD group treated with Ethosuximide, number, latens period and average duration of the reduction (p<0, 05). Omentin and Ethosuximide ctiveness of the DDD group (0, 05). Group B1(WAG/Rij) (n=8) was accepted as an control group and Group B2 (WAG/Rij) was used for Omentin experiment (p>0,05). As a result, Ethosuximide led to a decrease in the effective absance seizure.
