Abstract (EN)

In this research, molecular phylogenetic and taxonomic properties of Orchis anatolica BOISS. and Orchis tridentata SCOPOLI (Orchidaceae) taxa were investigated using the technique of sequencing of ITS1, 5.8S rDNA and ITS2 regions of genomic DNA.Genomic DNA isolation was performed from silica ? gel dried basal leaves and PCR technique was applied using with customized oligonucleotide primers for ITS1, 5.8S rDNA and ITS2 regions and also DNA sequencing process was completed with same primers. The data acquired from DNA sequencing were processed with some bioinformatic and phylogeny software and thus molecular taxonomy of these taxa were firstly became public. Alignment processes were performed with CLUSTALW2 software and constructing phylogenetic tree processes were performed with MEGA 4.0 (Stable) software using the methods of Neighbour ? Join (Saitou ve Nei, 1987).The data acquired from distance matrix table and phylogenetic tree were compared with classical taxonomic data and also phylogenetic relationships of O.anatolica and O.tridentata were discussed with other 17 Orchid taxa. With this thesis of master of science, it was observed that the phylogenetic tree which constructed with data acquired from ITS1, 5.8S rDNA and ITS2 regions of genomic DNA of O.anatolica and O.tridentata was compatible with classical taxonomy records. Whereas it was accomplished that ITS1, 5.8S rDNA and ITS2 regions were useful for molecular phylogeny and taxonomy studies with O.anatolica the ITS region doesn?t provide efficient data for O.tridentata taxa.
