Özet (EN)
In terms of growth area and production amount common bean have the first place in the world among the other granular leguminous seeds which are used for cooking. According to 2013 statistics, 21.365.119 tons in the world and 632.301 tons of beans are produced in our country. Beans as well as being an important food source it has importance for economic point and use in scientific research because of the animal can be used as feed and nitrogen fixation. Interest in organic agriculture has been increasing globally because of its advantages such as prevention of environmental pollution, biodiversity conservation, reduced fossil fuel consumption and more nutritious products. Although there are studies on nutritional value of organic products, extensive researches which are related to revealing the underlying molecular differences are almost negligible. In this study common bean ( Phaseolus vulgaris L.) genes which are differentially expressed in common bean grown under organic conditions compared to conventional plants were isolated. cDNA library was constructed from leaves of Phaseolus vulgaris L. Using Suppressive Substracted Hybridization (SSH) method. 94 clones were sequenced. Bioinformatic Analysis were performed to convert raw sequence data to EST data. The sequences were compared to the GenBank database of NCBI using BlastN, BlastP and BlastX tool. Blast results indicated that cDNA sequeces showed significant similarity to Fructose-bisphosphate aldolase, Rubisco, tRNA ligase, Hydrolase, Light regulated protein, F-box, ADP-ribosylation factor, Protein kinase, Tumor protein, Heat shock protein, Chlorophyll a/b binding protein, Glycine dehydrogenase protein encoding coding genes.
