Antiproliferative effects of essential oil and extract from Origanum vulgare L.

Abstract (EN)

Origanum species have been used extensively in pharmaceutical and food industry.In this thesis, aerial parts of Origanum vulgare were subjected to steam distillation to yield the essential oil. The compounds of essential oil were elucidated by GC-MS analysisAmong these compıunds,Cavracrol (90.4 %) was found as major product. In addition, plant material boiled during the steam distillation was filtered and water was evaporated under reduced pressure to yield the crude extract. Antiproliferative and Cytotoxic effects were investigated by BrdU and LDH techniques, respectively.A549 (human lung carcinoma),Hep3B (hepotocellular carcinoma),HT29 (human colon carcinoma), MCF7 (human breast adenocarcinoma) cancerous oils cell lines and FL (human amnion cells) normal cell lines were used.Cisplatin and 5-FU were used as standards. The essential oil revealedmore impactfulactivity against A549 (IC50, 27.2 µg/mL), Hep3B (IC50, 7.4 µg/mL), MCF-7 (IC50, 7.1 µg/mL) cancerous cell linesas compared to standards.In addition, the extract was observed to reveal outstanding activity on Hep3B (IC50, 27.2 µg/mL) and MCF-7 (IC50, 10.8 µg/mL) cell lines. The essential oil and extract have more cytotoxicity effect than the standards
