An effectiveness comparison of cabergoline and meloxicam to prevent ovarian hyperstimulation syndrome in a rat model

Abstract (EN)
Introduction: Ovarian hyperstimulation syndrome (OHSS) is a syndrome which have life threatening complications, emerging during ovarian stimulation to provide follicular maturation with gonadotropins at infertile women. The primary suspicious factor is vascular endothelial growth factor (VEGF) at this syndrome that depending on increased vascular permeability characterized clinic with ascites and pleural effusion. The other factors act also in pathogenesis of OHSS surely.Aim: A comparison of reduction of VEGF expression and efficiency of OHSS prevention by two drugs named Cabergolin and Meloxicam.Material and methods: We used immature Wistar female rats as experimental model of OHSS. A total of 50 rats were enrolled in the study and divided into five groups. Ten of them were divided as control group, mild OHSS were constituted in 10 of them and severe OHSS were constituted in remaining 30 of them that dividing into three equal groups. From rats that mild and severe OHSS were constituted by ovarian stimulation with gonadotropins, one of the severe OHSS group was given low dose 100 ?g/kg cabergolin (cb2) and the other was given 600 ?g/kg meloxicam. Groups were compared with each other in terms of body weights, vascular permeability, VEGF expression, ovarian weights and ovarian diameter.Findings: In our study, when control and mild OHSS groups were compared to severe OHSS group, manifest rising were determined in VEGF expression, vascular permeability, ovarian weights and ovarian diameters. It was shown that increase in VEGF expression was increased as a hCG dose dependent. But, it was shown that applied low dose cb2 and meloxicam treatments were not effective for reducing VEGF expression, vascular permeability, ovarian weights and ovarian diameters at severe OHSS.Conclusion: It was shown that VEGF increase have critical role on pathogenesis of OHSS. However, it was shown that applied treatments were not decreased VEGF expression and therefore, were not improved OHSS clinic.
