Investigation of the anti-cancerogenic effects of arum dioscoridis plant extract on pancreatic cancer cell line

Abstract (EN)

Pancreatic cancer, which is highly malignant, is a serious lesion with a high mortality rate due to the difficulty of early diagnosis and the risk of rapid metastasis. Phenolic compounds can be effective in many pathways in cancer initiation, progression, angiogenesis and metastasis stages. The aim of our study was to determine the anticarcinogenic effects of Arum dioscoridis ethanol extract (ADEE) on pancreatic cancer using PANC-1 cells in in vitro culture medium. Ethanol extraction of A. dioscoridis was performed. The total phenolic content of ADEE was determined as 2.4732±0.05 μg GAE/mg extract. The IC50 value was determined as 76.494 μg/ml by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical capture method to define the anti-oxidant activity. The highest amount of phenolic compound in the extract was determined as cynaroside (4,948 mg/kg) by LC-MS/MS. The effect on cell viability in PANC-1 and HEK293 cell lines was determined by MTT and the cells were treated with ADEE at IC50 doses for 48 hours. Western blot, quantitative expression analysis by qRT-PCR and immunoreactivity studies for inflammation detection were performed. As a result, it was observed that A. dioscoridis had anti-proliferative and anti-inflammatory effects in PANC-1 cell line, but no anti-cancer effect.
