Investigation of the effect of resvetratrol and avocado oil against paracetamol induced liver damage

Abstract (EN)

Paracetamol, also known as Acetaminophen and N-acetyl P-aminophenol (APAP), is also a derivative of Para-aminophenol. It is one of the agents commonly used in the clinic for analgesic and antipyretic purposes. Paracetamol intake at recommended therapeutic doses is generally well tolerated, while overdose causes toxicity. In this study, we aimed to investigate the protective effects of Resveratrol and Avocado Oil on liver damage induced by Paracetamol in rats. In our study, 5 groups (1. Group; Control, 2. Group; Paracetamol, 3. Group; Paracetamol + Resveratrol (RES), 4. Group; Paracetamol + Avocado Oil (AVO), 5th Group; Paracetamol + Resveratrol + Avocado Oil) was formed. Paracetamol was administered with a single dose of intraperitoneal injection at a dose of 600 mg / kg on the 4th day. Resveratrol 10 mg / kg and Avodako oil 200 mg / kg were administered to rats by oral gavage for 5 days. Animals were sacrificed at 24th hour following paracetamol administration. The liver tissue samples taken in our study were homogenized and TAS and TOS activities were determined spectrophotometrically. At the end of the experiment, liver damage was evaluated under light microscope by performing hematoxylin-eosin (H-E) staining on liver tissues of rats. In addition, the evaluation of DNA damage in blood was performed using the Comet Assay method. The results showed that TOS levels increased significantly in the paracetamol group compared to the control group. When the paracetamol group and RES, AVO and RES + AVO groups were compared, it was seen that the TOS level was significantly decreased. In addition, it was found that TAS levels in RES and RES + AVO groups increased significantly compared to the paracetamol group. The presence of necrotic areas was detected in the histopathological examination in the paracetamol group. It was determined that pre-treatment with RES and RES + AVO reversed oxidative stress parameters, DNA damage and necrosis caused by paracetamol. In this study, it has been shown that the combined use of resveratrol and avocado oil can provide protective effects against paracetamol-induced hepatotoxicity due to its antioxidant properties.
