Relationship between administration of paracetamol and hepatic free radical metabolism in rats: the effect of n-acetylcysteine

Abstract (EN)
In this study it was aimed to determine relationship between hepatic free radical metabolism and paracetamol (APAP) toxicity and effect of N-acetylcysteine (NAC). Purchased from GÜDAM those are weight 250-300 g thirty-six rats groupped into six as one control group. To control group only serum pyhsiological administered and to second group only was given 1000 mg/kg paracetamol. To third group was given only 400 mg/kg n-acetylcysteine. 1000 mg/kg paracetamol was given to fourth group 30 minutes before administration of 400 mg/kg NAC. To fifth group administered 1000 mg/kg paracetamol and after 30 minutes and 12 hours was given 400 mg/kg NAC. To last group was given 400 mg/kg NAC 30 minutes before administration of 1000 mg/kg paracetamol. SOD, CAT, GST, GSH-Px activities and AST, ALT, MDA levels were measured of each group. In conclusıon SOD activities were significiantly increased in third, fifth and sixth groups when compared to control group. CAT activities were also increased in fifth and sixth groups. Serum ALT values were significiantly decreased in fifth and sixth groups. There were no differences between groups MDA levels and GST activities. In histopatological evaluation, in the groups only paracetamol or only n-acetylcysteine administered hepatic injury was not seen. But in the groups n-acetylcysteine and paracetamol given together was seen injury.
