Protective effects of resveratrol against the damage caused by perfluorododecanoik acid on male rats

Perfluorododecanoic acid, a synthetic perfluorinated chemical containing 12 carbons, has broad industrial applications and has been detected in sera from humans and other animals and has been shown to produce adverse effects on the male reproductive system. In this study we aimed to investigate protective effects of resveratrol, against the damage Perfluorododecanoic acid on male rat histolochemically, imunohistochemically, ultrastructural and biochemically. 28 Sprague Dawley rats were divided into four groups. Control Group, no application was performed to this animals. Ethanol was given orally dosed for 14 days in sham group. Acid group, orally dosed for 14 days with 10 mg perfluorododecanoic acid. Resveratrol and acid group, orally dosed for 14 days with 40 mg and 10 mg perfluorododecanoic acid. Before the experiment and at the end of experiment, all rats were weighed and sacrificed on 14. days. Testes tissue slides were stained with hematoxylene eosine, Masson Trichrome and Periodic Acid Schiff for histochemically. TUNEL and Active Caspase 3 stains was performed to determine apoptosis immunohistochemically. Superoxide Dismutase, Glutathion, Malondialdehyde levels and serum testosterone were evaluated biochemically. Also ultrastructural alterations were observed. In histological evaluation, control and sham group?s testis tissues were observed normal. On acid group?s histochemically and ultrastructural investigations showed that seminifer tubules vacuolization, deprivation of spermatogesis cells and increase of connective tissue on interstitial area and thickened basale membran.No significant changes were seen in resveratrol and acid group which is protective effect of resveratrol. After immunohistochemical investigations apoptotic cell death was increased significantly in acid group compared with control group. In resveratrol and acid group, apoptotic cells decreased significantly compared with acid group.According to biochemically results Superoxide Dismutase, Glutathion, Malondialdehyde levels in acid group changed significantly. Acid group?s serum testosterone levels decreased significantly compare with the control and sham group. Resveratrol and acid group?s testosterone levels increased significantly compare with the acid and control group.In conclusion we assume that resveratrol can minimize the histological damage and biochemical differences significantly
