Abstract (EN)
Aim: At a time when new potential pathogens for periodontitis are emerging in the direction of developing technology, there is no reason not to expect bacteria that are culturally but not yet classified as pathogenic to be causative agents for periodontal abscesses. Many studies on periodontal abscess microbiology have been done on well-known bacterial strains and no study has been done on unknown bacterial strains that may be causative of the disease. In this study, it is aimed to isolate and identify the most intensively growing colony samples from aerobic, facultative anaerobic bacteria cultured on periodontal abscess specimens by using molecular methods. Material and Method: Periodontal abscess samples were obtained from 20 healthy volunteers who did not use any antibiotics during the last 4 weeks and were diagnosed as periodontal abscesses. The samples were then taken to the microbiology laboratory and were cultured. At the end of the second day, the numerically excess colonies were purified and stored at -86 ° C in NB (Nutrient Broth) containing 16% glycerol until further characterization. Then the genomic DNA was isolated. Each isolate was amplified from purified genomic DNA by 16S rRNA genes using PCR. After the obtained PCR products were visualized with a gel imaging system, DNA sequence analysis was performed through service intake. The BLAST procedure was performed to determine homologs with the nucleotide sequences NCBI libraries (http://blast.ncbi.nlm.nih.gov/blast.cgi) that were analyzed. As a result, at the end of the evaluations, the gene sequences were entered into GenBank and the acceptance numbers were obtained. Results: Sixty bacteria of 20 periodontal abscesses were isolated and diagnosed. Streptococcus species (S. pneumoniae, S. mitis, S. gordonii, S. anginosus, S. sanguinis, S. massiliensis, Streptococcus spp., S.cristatus, S. sinensis, S. mutans) in 33 samples, Neisseria species (N. Sicca, N. elongata, N.subflava) in 7 samples, Actinomyces species (A. naeslundii, A. odontolyticus) in 4 samples, Staphylococcus species (S. haemolytics, S. equorum, Staphylococcus spp.) , Morococcus cerebrosus in 2 samples and Moraxella spp., Bacillus spp., Enterococcus casseliflavus species in 1 sample. Conclusion: In our study, bacterial species detected were bacteria that caused abscesses and serious infections in different parts of the body, and these bacteria species were the first species isolated in periodontal apse for the first time. These bacteria, which are taken from the periodontal abscess content and show intense growth in the aerobic and facultative anaerobic environment, suggest that advanced studies for abscess microbiota are needed.
