Abstract (EN)

ELUCIDATION OF PHENOLIC COMPOUNDS OF THE PLANT Phytolacca americana (L.) BY HPLC-DAD Phytolacca americana (L.) belongs to the family of Phytolaccaceae and its motherland is North America. In this thesis study, the chromatographic separation and determination of the bioactive components of the this fruit were aimed. For this purpose, the methanol extract of the fruits was prepared. This extract was dried and dissolved with water and subjected to liquid-liquid extraction with diethyl ether, ethyl acetate, butanol respectively, resulting in four extracts together with the remaining aqueous part. Acidic hydrolysis was applied to the methanol extract so that the complex structures of the components could be disrupted and simpler structures could be determined. As a result of acidic hydrolysis, the aqueous portion was subjected to liquid-liquid extraction by sequential use of diethyl ether and ethyl acetate. These organic fractions were combined and another two-part extract was obtained with the remaining aqueous fraction. The methanol extract the six extracts from this extract were illuminated by a high performance liquid chromatography (HPLC-DAD) method combined with a diode-array detector. The order of the phenolic compounds quantitatively determined by HPLC-DAD analysis is ethyl acetate, butanol, hydrolysis organic, diethyl ether, methanol, water extract. Although there are similar components in the contents of the extracts, differences are observed between the extracts because they have rarely same components. In general, both flavonoid and phenolic acid contents of the extracts are rich. The extracts were found to contain p-OH benzoic acid, vanillic acid, citric acid, p-coumaric acid and derivatives, along with p-OH benzoic acid, vanillic acid, caffeic acid, ferulic acid and syringic acid. Among the flavonoids, rutin, quercetin, apigenin, luteolin and catechin derivatives were observed as well as rutin and quercetin.
