Abstract (EN)
Glu298Asp Polymorphism of the Endothelial Nitric Oxide Synthase Gene and Plasma Concentrations of Asymmetric Dimethylarginine in Pre-eclampsiaPregnancy is a physiological state same disorders occuring is pregnancy can pose negative effects to both mothers and fetus , the most promient one of the these negative effects is preeclampsia ,which is an important health problem of obstetric gynecology Insıdance of preeclampsia varies between 2 to 10 % in various nations since its physiopathology is not well establieshed this bring about the main difficulty in the treatment. Genetic factors nitric oxide (NO) dependent vasodilatation play important role in its development. In the current research, it was aimed to investigate whether or not genetic polymorphism of endotelial nitric oxide synthase (eNOS) can play a role in preeclampsia and to enlighten the correlation of Asymmetric dimethylarginine (ADMA) accumulation with this hypotesis. G/A polymorphism encoding Glu 298Asp variant was studied. Polymorphism of this gene cause alteration in protein structure and is related to NO synthesis . Glu 298Asp variants, ADMA and arginine levels were composed pregnant women with and without preeclampsia. 55 women with preeclamptic pregnancy and 54 normal pregnant women were included in study. The difference in eNOS Glu 298Asp genotype distribution and allel frequences of preeclampsia and normal pregnant women were found to be statistically insignificant . ADMA,SDMA , arginine and ADMA /SDMA ration were found to be significantly higher in preeclampsia pregnant women composed to women with normal pregnancy (p<0.0001) . In women with normal pregnancy arginine /ADMA ratio was found to be increased composed to these with preeclamptic pregnancy , however this difference was statisticlly insignificant .In conclusion , no direct effect of eNOS Glu 298Asp Polymorphism on preeclampsia risk was found .İt appears that ADMA ,SDMA and arginine measurements could be benificial in determining preeclampsia development .Key words : ADMA, eNOS gene polymorphism, preeclampsia
