Abstract (EN)

Alzheimer's disease (AD) is a disease that develops as a result of the accumulation of soluble amyloid species as insoluble amyloid plaques, leading to neuroinflammation in nerve cells and dysfunctions in cholinergic synapses by increasing oxidative stress and manifests itself with progressive cognitive impairment. Today the disease The fact that the treatment can only be applied symptomatically with a limited number of drugs makes the discovery of new drug molecules important. The fact that the most effective drugs used for AD are of herbal origin (Galantamine, Ginkgo biloba leaf extract, etc.) Considering this, plants are thought to be an important resource in the search for new drug molecules. From this point of view, it is extremely common in our country and has internal use, especially as an anti-inflammatory, and also; antioxidant and anti-inflammatory properties, especially rosmarinic acid In this study, the role of Prunella vulgaris L. plant, which contains many phenolic acids and has been the subject of some studies, in the treatment of AD will be investigated by in vitro methods. Since it is known that oxidative stress and inflammation developing in nerve cells due to amyloid beta (Aβ) accumulation play an important role in the pathophysiology of AD, the activities of aqueous ethanolic extract of P. vulgaris plant, fractions rich in effective compounds and isolated pure compounds will be examined through these mechanisms. For this purpose, firstly, aqueous ethanolic extract of the aerial parts of P. vulgaris plant on AH in vitro. effects will be investigated and then the compounds thought to be responsible for the activity will be isolated in pure form by various chromatographic methods in order to find drug candidate molecules effective against the disease. The in vitro activities of the extracted and isolated pure compounds will be investigated in terms of the effects of the samples on cell viability and thus their protection against the negative effects of Aβ25-35 plaques by the MTT method in SH-SY5Y (human neuroblastoma) cells stimulated with Aβ25-35 peptide. In addition; Acetylcholinesterase and butyrylcholinesterase inhibitory effects of extracts and pure compounds will also be investigated in vitro. This thesis has important results in terms of finding a new herbal product or drug candidate molecules with high added value that can have serious contributions to AD. will be able to provide.
