Abstract (EN)

Psoriasis is a chronic, relapsing, sharply demarcated inflammatory skin disease which is characterized by bright nacre squamous on plaque or papules. The incidence worldwide varies depending on ethnic, geographical and environmental factors and can affect men and women of all ages. Both genetic and triggering factors are involved in the etiology of the disease. The features of psoriasis lesions are in the form of inflammatory cell infiltration, hyperproliferation of keratinocytes, and tissue inflammation consisting of T lymphocytes in the epidermis and dermis. These changes occur as a result of activation of the immune system. Cytokine signal suppressors (SOCS) are eight intracellular proteins families that act as endogenous inhibitors of proinflammatory pathways triggered by various cytokines. Cytokines are structurally interrelated and play a role in regulating immunity and inflammation. SOCS proteins are inhibitors of the cytokine signal and their expression is induced by the JAK/STAT pathway. To our knowledge, there are no studies investigating the association between SOCS gene polymorphisms and psoriasis. The study included 100 unrelated patients diagnosed with psoriasis and 100 healthy individuals without any inflammatory disease. The SOCS3 rs4969169 and SOCS7 rs3748726 polymorphisms were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. SOCS3 rs4969169 genotype frequency; CC and CT genotypes were 67%, 33% in the patient group; they were 73%, 27% in the control group. SOCS7 rs3748726 genotype frequency; TT, TC and CC genotypes were 89%, 9%, 1% in the patient group; they were 91%, 8%, 1% in the control group. The polymorphisms of SOCS3 rs4969169 and SOCS7 rs3748726 were found to have no effective role in the pathogenesis of psoriasis. As it is the first study to investigate the effect of rs4969169 and rs3748726 in psoriasis, it would be beneficial to repeat the study in different ethnic populations with more patients and healthy individuals.
