The role of the Granulysin in the pathogenesis of psoriasis

Psoriasis etiology is an autoinflammatory dermatosis characterized by sharply defined, erythematous, pearlescent white squamous cells of unknown origin. The most accepted mechanism in the etiopathogenesis of psoriasis is the induction of inflammation with keratinocyte hyperproliferation. Granulizin is a cytolytic antimicrobial peptide (AMP) that is secreted together with granzyme and perforin from the granules of human cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, it has been immunohistochemically proven that the expression of granulisin is increased in lesions of psoriasis. However, there is not investigating the relationship between GNLY gene polymorphisms and psoriasis. 100 unrelated patients diagnosed with psoriasis and 100 healthy individuals without any inflammatory disease were included in our study. GNLY rs7908 and GNLY rs10180391 gene polymorphism frequencies were determined using PCR and RFLP method and the results were analyzed by chi-square and logistic regression analysis. GNLY rs7908 genotype frequency was 51%, 41% 8% for GG, GC and CC genotypes in the patient group; 37%, 44% and 19% in the control group. GNLY rs10180391 genotype frequency was 41%, 47%, 17% for CC, CT and TT strains in the patient group; 44%, 41%, 15% in the control group. We found that the genotype of GNLY rs7908 CC and the C allele showed a protective effect against psoriasis and reduced the severity of the disease that the rs10180391 SNP did not play an effective role in the pathogenesis of psoriasis. Functional studies of GNLY rs7908 and GNLY rs10180391 gene polymorphisms on the risk of developing psoriasis and on disease progression will lead to the development of new treatment methods against psoriasis.
