Determination of antioxidant capacity ve activity on some metabolic enzymes of puerarin compound in vitro conditions

Abstract (EN)

In the thesis study, the inhibition effects of puerarin compound with a phenolic molecule structure on acetylcholinesterase (AChE) ve butyrylcholinesterase (BChE) enzymes were investigated. While the IC50 value obtained for AChE enzyme was 177.73 nM, it was calculated as 315.07 nM or BChE. Tacrine was used as the stveard molecule. It was determined that it showed more effective inhibition than the tacrine molecule. In the other part of the thesis, antioxidant activity was evaluated for the same molecule. For this purpose, determination of Fe3+-Fe2+ reducing capacity, 2,2-Azino-bis (3-ethylbenzo-thiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, Cu2+-Cu+ reducing capacity (CUPRAC method), 1,1-Diphenyl 2-picryl hydrazil (DPPH) free radical scavenging activity and DMPD radical removal method were used. BHT, BHA, ascorbic acid (vitamin C), trolox ve α-tocopherol (vitamin E) were taken as reference antioxidants ve compared. For ABTS radical scavenging activity, the % inhibition amount of puerarin molecule at 20 mg/mL concentration was calculated as 99.33%. It was determined that the reduction capacities were in parallel with the stveards. It is thought that the results obtained will shed light on cosmetics, drugs, medical treatments ve applications, ve new biochemical studies.
