Abstract (EN)

MolecularyDetection Of RifampcinResistanceInMycobacteriumtuberculosisStrainsIsolatedFromSputumSamples Of PulmonaryTuberculosisPatients Resistancetorifampicin (RIF) cause a seriousandsignificantthreattotuberculosiscontrol. Control of drugresistanttuberculosisreliespropertreatment of drugresistantcasesandrapiddiagnosis. Molecularcharacterization RIF-resistantM. tuberculosisclinicalstrains of differentorigins can generateusefulinformationforMDR and XDR epidemiology. Theaim of thisstudywastoinvestigatethemolecularepidemiology of rifampicinresistantMycobacteriumtuberculosisstrainsisolated in CukurovaRegion, SouthernTurkey. Weevaluatedthemutationsin a 327basepairslocated in 481-589 codons of therpoB gene byautomatedsequencing of 50 strains. In 50rifampicinresistantstrains, mutationswereidentified in 98% strains. No mutationwasdetected in the(%2) 1resistantstrains. Themostcommonmutation (51,02%) was Ser531Leu. Overall, 15 differentmutationsaffecting 12 codonswereidentifiedThecodonnumbers of themostfrequentlyencounteredmutationswere 531 (65,30%), 516 (10,20%), and 526 (6,1%). Itwasconcludedthattheresultsfromthisstudy can serve as a basisformonitoringmolecularepidemiology of drugresistantM. tuberculosisstrainsisolated in ourregion.
