Effect of glutamine and argine treatment on inflammatoryresponse in rats with pulmonary contusion model

Abstract (EN)
Purpose: In our study; We evaluated histopathologically whether glutamine and/or arginine treatment, which has an important role in the immuno-inflammatory response, is effective in the experimental trauma model. The aim of our study is to investigate the effectiveness of glutamine and/or arginine treatment in the early period for the reduction of inflammation after pulmonary contusion. Method: In our study, 24 adult female Wistar Albino rats with an average weight of 460 g were used from the same colony. Rats were randomly divided into five groups; They were randomly grouped as 3 in the sham and trauma groups, and 6 in the arginine, glutamine and glutamine+arginine groups. While trauma was not applied to the rats in the first group, contusion was applied to the other group of rats with the modified form of the isolated bilateral pulmonary contusion model described by Raghavendran et al. The rats in the first group did not receive any treatment, the rats in the second group were administered intraperitoneally (i.p) 0.9% saline. The rats in the third group were treated with i.p glutamine, the fourth group rats with i.p arginine, and the rats in the last treatment group were given i.p. glutamine + arginine treatment. Sacrification was performed three days after the procedure in all groups. Histopathological evaluation was performed after sacrification. Three different parameters in histopathological evaluation; Inflammation intensity, intraalveolar hemorrhage and alveolar congestion were examined histopathologically. Results: In the mutual analysis of glutamine and arginine groups with trauma; In both glutamine and arginine groups, improvement was observed in all three histopathological parameters and statistical significance was determined. As a result of the individual administration of glutamine and arginine in rats, similar effects were observed in both molecules. In the evaluation of the treatment subgroups, 4 although the combined administration of glutamine and arginine in the treatment showed similar effects in terms of intraalveolar hemorrhage and alveolar congestion compared to glutamine, it was observed that it caused more negative results in terms of parenchymal inflammation intensity than single glutamine treatment. In all three scoring; It was observed that glutamine and arginine treatment were less effective than arginine treatment alone in rats. Conclusion: In the treatment of inflammation after pulmonary contusion; Although beneficial results were observed with the separate administration of glutamine and arginine, we found that the combined administration of these two molecules was less effective compared to the other treatment groups.
