Abstract (EN)
Puprose: Nephrotic syndrome is a disease that may give rise to the serious morbidities after childhood period. Several treatment procedures are used, from steroid treatment to immunosuppressants. Antiproteinuric and renoprotective effects are tried in several medicines. In our study, we aimed to investigate the effect of 1,25 (OH) 2D3 vitamin and paricalcitol proteinuria, renal tissue histology, podocin and nephrin upon mRNA. Materials and Methods: 28 number of rats has been divided into 4 groups. Experimental nephrotic syndrome has been provided with 10mg/100gr of puromycin. Group 1 has been determined as a healthy control group, Group 2 as a nephrotic syndrome group formed by PAN, Group 3 as a vitamin D group, Group 4 as a paricalcitol group. No medication has been realized to Group 1. Only puromycin application has been performed to Group 2. 1,25 (OH) 2 D 3 application has been realized intraperitoneally to Group 3 in dose of 0,4 µg/kg/day every day for 7 days after performing the application of puromycin. Paricalcitol application has been realized intraperitoneally to Group 4 in dose of 240ng/kg/day every day for 7 days after performing the application of puromycin. Spot urine samples have been collected after performing massaged on the 0th, 4th and 7th days. After taking renal tissue from the rats after 7 days, rats have been sacrificed by cervical dislocation method. Results: An increase of urine protein/creatinine ratio in nephrotic syndrome within itself has been found significant (p=0.015). In addition, increase of protein/creatinine within days is significant also in groups of paricalcitol and vitamin D (p=0,008,p:0,030). However, when compared between groups, the presence of a statistically-significant difference between urine protein/creatinine ratio on 0th day makes the comparison between the groups meaningless in the following days (p=0,028). No significant difference could have been detected between the groups in mRNA expression of nephrin (p=0,721). In mRNA expression of podocin, no statistically significant increase has been detected according to the nephrotic syndrome group (p = 0.003). Glomerular hypersolarity has been found out in the nephrotic syndrome group in performed histological studies. And minimal glomerular hypersolarity has been detected in paricalcitol and vitamin D group. However, no significant immunoreactivity could have been detected in the immunohistochemical studies realized for podocin and nephrin (p= 0,356, p= 0,475). Conclusion: As a result, under the light of the studies which have been performed with paricalcitol and Vitamin D and theoretical information, we are aware that there are antiproteinuric and renoprotective effects. Although we have observed numerically this effect in the study that we have carried out, statistical significance has been achieved.
