Özet (EN)

Effects of Vascular Endothelial Growth Factor (VEGF) on Developing Organ Damage During Experimental Intestinal Ischemia Reperfusion Injury Model in RatsWhen blood flow is not enough for cellular functions ischemia occurs. Ischemia-reperfusion injury of the intestine is an important factor associated with high rates of morbidity and mortality. Paradoxically, restoration of blood flow to the ischemic tissue initiates a cascade of events that may lead to additional cell injury known as reperfusion injury.Vascular endothelial growth factor (VEGF) is the angiogenic factor promoting processes of neovascularization taking place in the embryo and the adult. VEGF is required to sustain newly formed vessels and plays additional multiple roles in the maintenance and function of certain mature vascular beds.In this study Wistar Albino male rats were used. Rats were divided into five groups (n= 6). Groups: Sham, VEGF, I/R, I/R-VEGF, VEGF-I/R. In sham and VEGF groups only surgical procedure is applied. In I/R and VEGF groups, surgical procedure and clip is applied on the mesenteric artery for 90 min. For I/R-VEGF and VEGF-I/R groups, before or after than ischemia period one dose intra venous VEGF (0,8 ?g/kg) was given. 4 hours after than ischemia period, blood and tissue samples were collected. All procedure were done under anaesthesia. On the tissues biochemical and immunohistochemical methods were used for analysis. Blood samples were taken for biochemical analysis.Biochemical analysis measurement results are correlated with the results obtained from histological intestigations. The differences are determined between the groups by using Mann Whitney U tests. Our findings showed that VEGF, which is given before than ischemia, has most protective effects on intestinal ischemia-reperfusion injury. Keywords: Ischemia-Reperfusion, Intestinal Ischemia-Reperfusion Injury, Oxidative Stress, Free Radicals, Antioxidants.
