2. ABSTRACTEffects of Erdosteine on inflammation and fibrosis in rats with pulmonaryfibrosis induced by BleomycinIdiopathic pulmonary fibrosis (IPF) is an interstitial pulmonary disease. Theetiology and the pathogenesis of fibrosis and alveolitis are unknown. Current treatmentmodality is also poor for this condition.In this experimental study, pulmonary fibrosis was obtained. Some chemokineslevels, inflammatory cell counts were measured in broncho-alveolar lavage (BAL) fluidand pulmonary tissue specimens. Thus, these agents were investigated to understandtheir role in the pathogenesis of IPF and the role of Erdostein on acute inflammatoryresponse and fibrosis was aimed to explore in this study.Wistar type male rats were taken into the study. Intra tracheal (i.t.) saline wereadministered in day 0 in control group (n=15). Bleomycine (BLM) were given i.t. tobleomycine group (7,5 U x kg-1) (n=15) and erdostein was given orally (10 mg x kg-1 xday-1) after BLM (7,5 U x kg-1) application to erdostein group (n=15). Five rats weresacrificed in each group to obtain BAL fluid in day 0, day 14 and day 29. MDA, MIP-1α and MIP-2 levels in BAL fluid, inflammatory cell counts, hydroxy-proline levels inlung tissue were measured. Histopathological examination were also performed.MIP-1α, MIP-2, MDA levels in BAL fluid were statistically significant reducedby erdostein in erdostein group compared in BLM group in acute inflammatory phase(day 14) and late fibrosis phase (day 29). (P levels for neutrophils were 0.008 and 0.009,for MIP-1α were 0.016 and 0.009, for MIP-2 were 0.009 and 0.016, for MDA were0.009 and 0.009 in day 14 and day 29, respectively.) Fibrosis levels were foundsignificantly lower in erdostein group than BLM group in day 29 (p=0.005).In conclusion, it is suggested that erdostein may have protection affect BLMinduced fibrosis and acute pulmonary inflammation via suppression of neutrophilsaccumulation, inhibition of lipid peroxydation and inhibition of chemokines productionand release.Key words: IPF, MDA, MIP-2, Hydroxyproline, Erdostein.
