Investigation of the effects of vinpocetine on the liver in sepsis created by cecal ligation and puncture method in rats

Abstract (EN)

Introduction: It is known that vinpocetine, which is a vinca alkaloid and effects by inhibiting the phosphodiesterase (PDE) enzyme, has various pharmacological effects and studies continue in this area. However, there is no study in the literature on the effects of vinpocetine on liver damage in sepsis. In this study, we aimed to investigate the effects of vinpocetine on liver damage in a rat model with sepsis. Materials and methods: In the study, 28 Wistar albino female rats were randomly divided into 3 groups; The SHAM group (n=8) was used as the control group, the CLP group (n=10) was the group in which sepsis was created, and the sepsis was created by the CLP (cecal ligation puncture method). After the administration of 5 mg/kg vinpocetine for 10 days to the VINPO+CLP (n=10) group, sepsis was induced by the CLP method. After the weight and body temperature measurements, complete blood count, serum aminotransferases, lactate, albumin, glucose, lactate dehydrogenase (LDH) and creatine cinase (CK) were studied. Histopathological examinations were performed in the liver tissue, and also biochemically in the liver tissue, Interleukin-1 beta (IL-1β), IL-6, Tumor Necrosis Factor - Alpha (TNF-α), Nuclear Factor-Kappa B (NF-Κb), Glutathione (GSH), Malondialdehyde (MDA), Total Oxidant Capacity (TOS), Total Anti-Oxidant Capacity (TAS) and Oxidative Stress Index (OSI) levels were studied. Results: It was observed that serum ALT, AST, LDH, CK, and MDA and IL-1β, which were studied in tissue biochemistry, were significantly decreased in the group given vinpocetine compared to the group not given vinpocetine. Smaller infiltration areas were detected in the histopathological sections of the group given prophylactic Vinpocetine. Conclusion: These findings show that vinpocetine may be protective from liver damage in sepsis with its possible antioxidant and anti-inflammatory effects.
