Abstract (EN)

In this study, it was aimed to evaluate the effects of orally administered resveratrol on bacterial translocation and inflammation in an experimental peritonitis model induced by cecal perforation. A total of 30 male Wistar-Albino rats, aged 12 months and weighing between 380–625 grams, were used. The rats were randomly divided into three groups: Sham (n=10), Control (n=10), and Treatment (n=10). All rats underwent laparotomy under anesthesia, and the cecum was isolated to expose the ileocecal valve. In the Sham group, no additional procedure was performed; the abdomen was closed 8 minutes after laparotomy. In the Control and Treatment groups, the cecum was ligated distal to the ileocecal valve, and cecal ligation and puncture (CLP) was performed by two punctures using an 18-gauge needle from the antimesenteric surface. The abdomen of the Control group was then closed without any treatment. In the Treatment group, 100 mg/kg of resveratrol suspended in CMC was administered orally by gavage following the CLP procedure. Eight hours after the surgical intervention, all rats underwent relaparotomy under anesthesia. Blood samples were collected from the inferior vena cava, and the rats were sacrificed by surgical exsanguination. Tissue samples were obtained from the liver, spleen, and mesenteric lymph nodes (MLN), and blood samples were taken for culture. Inflammatory markers (TNF-α and IL-6) were measured from serum using ELISA. Bacterial translocation was evaluated by culturing blood and tissue samples, and results were expressed as colony-forming units (CFU) per gram. This study suggests that oral resveratrol administration may reduce systemic inflammation and limit bacterial translocation in an experimental peritonitis model.
