Effects of regular and continuous exercise on oxidative stress and apoptosis in rats

Abstract (EN)
In this study, it was aimed to investigate the effect of regular and continuous exercise on the oxidative stress parameters and apoptosis related Bax and Bcl-2 protein levels with acute or intense acute exercise in rats. For this purpose, 5 groups of 35 Wistar-Albino male rats were formed, with 7 in each group. While no application was made to the control group, the other four groups were exercised at a speed of 1.5 km / h for 1 hour on a 20 degree incline treadmill every day. Control and Group 1 were sacrificed under anesthesia at the end of Day 1, Group 2, Day 7, Group 3, Day 15 and Group 4 at day 30. Skeletal muscle and blood samples were taken from the groups, malondialdehyde (MDA), glutathione (GSH) levels, catalase (CAT) and glutathione peroxidase (Gsh-Px) enzyme activity spectrophotometrically, Bax and Bcl-2 protein expression levels, which play a role in apoptosis, were evaluated by western blot technique. According to the results obtained, when the exercise groups are compared with the control group, there was a significant increase (P <0.05) in skeletal muscle MDA levels in the 1st and 2nd groups, and a significant increase in plasma MDA values in the 1st group (P <0.05). It was observed. There was no difference between the control group and the other exercise groups. In the skeletal muscle and plasma GSH-Px values, the increase in the 4th group was found to be significant compared to the control group (P <0.05). When the skeletal muscle CAT values were examined, the increase in the 2nd, 3rd and 4th groups was found to be significant compared to the control group (P <0.01). When the plasma CAT values were compared with the control group, the increase in the 4th group was found to be significant (P <0.05). When Bax and Bcl-2 protein expression levels were compared with the control group, Bax protein expression level increased in group 1, Bcl-2 protein expression level decreased and Bax / Bcl-2 ratio increased (p <0.05), when the 2nd, 3rd and 4th groups were compared with the control group, there was no difference between Bax and Bcl-2 protein expression levels and Bax / Bcl-2 ratio (p> 0.05). As a result, it was concluded that regular and continuous exercise has a protective effect against oxidative stress and apoptosis triggered by acute exercise can be suppressed by regular and continuous exercise.
