Effect of topical curcumin-cyclodextrin complex onhealing in the treatment of second degree burns in rats

Abstract (EN)
Introduction and purpose: This study aimed to investigate the effects of topical cyclodextrin complex of curcumin in partial burn treatment in rats. The main problem of the study is that there is no effective and economical treatment option that will accelerate wound healing, reduce the need for early tangential excision, reduce necrosis and minimize side effects in deep second degree burn treatment. The most commonly used topical silver sulfadiazine treatment for second-degree burns has been restricted due to its side effects. The main hypothesis of the study is that curcumin, which has anti-inflammatory, antimicrobial, antioxidant, antiischemic and proangiogenic properties, can be used as a reliable source in burn treatment by increasing its effectiveness with cyclodextrin. Materials and Methods: The study was conducted at Erciyes University DEKAM. Rats (Rats) weighing 200-400 g, with ages ranging from 4 to 6 months, were included in the study. After statistical power analysis, 40 rats were randomly divided into four groups, 10 rats in each group. During the experimental process, rats were fed in separate cages and kept at a constant room temperature of 21°C and 45±10% humidity by meeting their standard water needs. 10 animals were selected for each group in rats, but two animals in each group died after anesthesia was given for the burning process and the study was completed with 8 animals in each group. The wound size in rats was adjusted as 2 pieces of 2cm x 2cm on a rat's back. Anesthesia was provided by giving 50 mg/kg ketamine and 15 mg/kg xylazine while creating the burn and taking tissue samples in rats. Results: According to the intragroup comparison results between the wound and control wound sizes according to the 3rd, 7th, 14th and 21st day biopsy dates of all subjects in all groups; In all wound area biopsies (wound sizes on days 3, 7, 14 and 21), the wound sizes of the subjects in Group 2, namely the Curcumin-cyclodextine complex group, were reduced statistically significantly more compared to the wound sizes of the subjects in Group 1 (Sham group), Group 3 (Curcumin group) and Group 4 (Cyclodextine group). While the difference between the wound sizes of the subjects in Group 2, namely the cyclodextrin complex of curcumin group, and the control wound sizes were not statistically significant on the 1st biopsy date, the wound size was reduced statistically significantly more compared to the control wound size on the 2nd, 3rd and 4th biopsy dates. The wound size measured on the 4th biopsy date of the subjects in the curcumin-cyclodextine complex group was reduced statistically significantly more compared to the control wound size on the 1st biopsy date Conclusion: Cyclodextrin complex of curcumin provides more effective results in the treatment of 2nd degree burns than Curcumin and Cyclodextine used alone Keywords: 2nd Degree Burn, Wound Healing.
