Effects of the intrathecally administered ketorolac trometamin on spinal cell injury in trauma model of medulla spinalis in rats

SUMMARYEFFECTS OF THE INTRATHECALLY ADMINISTERED KETOROLACTROMETAMIN ON SPINAL CELL INJURY IN TRAUMA MODEL OF MEDULLASPINALIS IN RATSObjective: The aim of this study was to investigate the effects of intrathecallyadministered ketorolac trometamin on spinal cell damage, in spinal cord traumatizedrats.Material and Method: Eighteen rats were included in this study. The spinal cord wastraumatized, after spinal catheter was placed and fixed to the each rat underanesthesia. The subjects were randomized into three groups: Group S (n:6); Sterilesaline solution (10 µL) was administered by spinal catheters; Group K50(n:6); Fiftymicrogram ketorolac trometamin solution was diluted in 10 µL of saline and wasadministered to subjects through spinal catheters; Group K400 ( n:6); Four hundredmicrogram ketorolac trometamin was diluted in 10 µL of saline and was administeredto subjects through spinal catheters. The spinal catheters were flushed with 5 µL ofsterile saline solution after the drug administration in all groups. The scores accordingto the ?Behavior Rating Scale (BRS)? were determined at the 4th, 12th, 24th, 48thand 72nd hours after intrathecally administration of ketorolac tromethamin. Thecross sections of spinal cord were examined pathologically. The percentage of thedamaged spinal cord area was determined in cross sections.Results: When groups were compared according to the BRS scores; the meanvalues of the BRS scores was significantly high in group K400 (p < 0.05) at 48th and72nd hours. The mean values of the damaged spinal cord area were significantly lowin group K50 and group K400 (p<0.05). The mean values of the histopathologyscores were compared and it was significantly low in group K400 (p< 0.05).Conclusion: In this study, intrathecally administration of 400 µg ketorolactromethamine resulted in the attenuation of the spinal cell damage in spinal cordtraumatized rats.Keywords: intrathecally, ketorolac trometamin, spinal trauma, spinal cell damage
