The effect of ozonated oil on experimentally induced skin infection BY Streptococcus pyogenes and staphylococcus aureus in rats

Abstract (EN)

The aim of the present study was to determine the effect and activity of ozonated olive oil on experimentally induced skin infection by Streptococcus pyogenes and Staphylococcus aureus in rats. In this study 93 female Wister Albino rats aged between 6-8 weeks and weighing 80-130 gr were used as animal material. The study was performed on 3 main groups which Group 1. was infected with Streptococcus pyogenes Group 2. was infected with Staphylococcus aureus and Group 3. was negative control group. Infection were made in the skin of the experimental animals used in this study. Furthermore, each of Group 1. and Group 2. were further divided into 4 subgroups. Apart from Streptococcus pyogenes infection, animals in Group 1.1. (n=10), received ozonised olive oil (Aktifoks, Işık Kozmetik), animals in Group 1.2 (n=10) received olive oil, Group 1.3. (n=10) received fusidic acid (Stafine cream, Koçak Farma İlaç) to the infection side in their skin at 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours. Animals in Group 1.4. (n=11) did not received any treatment after Streptococcus pyogenes infection. Swab and tissue samples before treatment and 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours after treatment were collected and evaluated from Groups 1.1., 1.2., 1.3. and 1.4.. Apart from Staphylococcus aureus infection, animals in Group 2.1. (n=10), received ozonised olive oil (Aktifoks, Işık Kozmetik), animals in Group 2.2. (n=10) received olive oil, Group 2.3. (n=10) received fusidic acid (Stafine cream, Koçak Farma ilaç) to the infection side in their skin at 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours. Animals in Group 2.4. (n=11) did not received any treatment after Streptococcus aureus infection. Swab and tissue samples before treatment and 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours after treatment were collected and evaluated from Groups 2.1., 2.2., 2.3. and 2.4.. Abraded skin regions in Group 3. (n=11) which was negative control group, received only serum physiologic at 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours. Furthermore, swab and tissue samples before treatment and 24, 28, 32, 40, 48, 56, 64, 72, 80, 88, 96 hours after serum physiologic application were collected and evaluated from Group 3.. In histopathological examination, as a result of gram staining, in Group 1.1. (ozonized olive oil group) presence of bacteria was 18.2 % and in Group 1.4. (infected control group) the presence of bacteria was 100 % which ozonized oil caused significant (p<0.001) reduction in the presence of bacteria compared to infected control group (Group 1.4). In addition Group 2.1. (ozonized olive oil group) presence of bacteria was 45.5 % and in Group 2.4. (infected control group) the presence of bacteria was 100 % which ozonized oil caused significant (p<0.001) reduction in the presence of bacteria compared to infected control group (Group 2.4.). Furthermore, with concern to dermal lesions, when presence of inflammatory cells compared it was higher (p<0.001) in Group 1.4. compared to Group 1.1.. Additionally, with concern to dermal lesions, when presence of inflammatory cells compared it was higher (p<0.001) in Group 2.4. compared to Group 2.1. But, statistical importance were not observed only in papillary dermis in both Group 1. and Group 2.. When groups were compared with concern to incrustation and recovery; between ozone group (Group 1.1.) and infected control group (Group 1.4.) at 40, 48, 56, 64, 72, and 80 hours after infection, incrustation and recovery were better (p<0.01) in Group 1.1. compared to Group 1.4. In addition, When groups were compared with concern to incrustation and recovery; between ozone group (Group 2.1.) and infected control group (Group 2.4.) at 40, 48, 56, 64, 72, and 80 hours after infection, incrustation and recovery were also better (p<0.01) in Group 2.1. compared to Group 2.4.. As a result, in the present study, experimental skin infections with Staphylococcus pyogenes and Streptococcus aureus were made and the effects of ozonized olive oil in the treatment of the infection were evaluated and results were compared with other treatment methods. After occurrence of the infection 0.1 gr ozonized olive oil had anti-microbial and anti-inflammatory effect and observed to have healing activity at 72nd and 80th hours after infection. Furthermore, the effects of ozonized olive oil were comparable with the effects of Fusidic acid which is well known and its activity has been proven in the skin infections with Staphylococcus and Streptococcus. Additionally, as far as known microorganisms has no resistance development against ozone and has no important side effects.
