Protective effect of naringenin in renal ischemia-reperfusion injury in rats

Background/Aim: To prevent structural, functional deterioration in renal tissue due to ischemia-reperfusion injury, we aimed to investigate protective, therapeutic effects of naringenin, a natural flavonoid, in rat kidney tissue from biochemical and histological point of view. Materials/Methods: 32 Wistar albino female rats were divided into 4 groups as sham, IR, NRG/IR, IR/NRG. Sham group underwent right nephrectomy. Other groups, after the right nephrectomy, the left renal pedicle was clamped, renal damage was induced by 60 min ischemia and 24 hr reperfusion. In the NRG/IR, IR/NRG groups, 100 mg/kg i.p. NRG was applied. End of the application under anesthesia; Blood samples, left kidney tissue were taken by intracardiac route. Serum BUN, creatine, albumin; MDA, SOD, GSH, CAT, GSH-Px, were measured in tissue samples. Blc-2, Bax, kisspeptin were examined for light microscopy, apoptotic evaluation. Key words: renal ischemia-reperfusion, rat, naringenin, kisspeptin, Bcl-2,BAX. Results: Decreasing enzymes such as SOD, CAT increased in NRG/IR, IR/NRG groups. In the IR group, MDA, NRG/IR, IR/NRG groups decreased. BUN, creatine increased in the other groups compared to the sham group. There was a significant increase in Bcl, in IR/NRG, NRG/IR groups compared to other groups. Kisspeptin staining; IR group decreased compared to sham group, IR/NRG group increased compared to IR group. Conclusions: Ischemia reperfusion injury disrupts oxidant-antioxidant balance, causing structural, functional damage to kidney tissue and even kidney loss. NRG has a protective and therapeutic effect on kidney tissue damage due to its antioxidant, anti-inflammatory properties.
