Investigation of synergistic cytotoxic effects of rimcazole and p-coumaric acid, o-coumaric acid, curcumin combinations on Caco-2, A549 and 3T3 cells

Abstract (EN)

Today cancer is becoming an increasingly important health and life problem. According to research, it is expected that the number of new cases in the world will be 27 million in 2030, 17 million deaths due to cancer and the total number of cancer patients will be 75 million. In this thesis, it is aimed to determine the synergistic/additive cytotoxic effects of rimcazole, which is a selective sigma-1 receptor antagonist and considered to be anticancer, and rimcazole's combinations with p-coumaric acid, o-coumaric acid and curcumin in Caco-2, 3T3 and A549 cells. For this, cytotoxic effects of single and combination applications of rimcazole, p-coumaric acid, o-coumaric acid and curcumin in Caco-2, 3T3 and A549 cells were investigated by MTT method and coefficient of drug interaction (CDI) values were calculated in each cell. The cytotoxic effects of rimcazole and, rimcazole with p-coumaric acid, o-coumaric acid, curcumin in Caco-2, 3T3 and A549 cells occur depending on concentration. According to the CDI calculation results due to cell viability; it has been determined that combined applications rimcazole combined with curcumin have a strong synergistic effect (CDI < 0.7) on A549 cells at a concentration of 12.5 µM, rimcazole combined with o-coumaric acid have a strong synergist effect (CDI < 0.7) on both 3T3 and A549 cells at a concentration of 25 μM, rimcazole combined with p-coumaric acid have a strong synergistic effect (CDI < 0.7) on A549 cells at a concentration of 12.5 µM.
