Investigation of the Potential Therapeutic Effect of Rutaekarpin on Metastatic Prostate Cancer Cell Lines INTRODUCTION AND OBJECTIVE: Prostate cancer with an increased incidence in the world and Turkey is one of the public health-threatening malignancies. In particular, metastatic prostate cancer is an important cause of death in men despite the combined use of more than one chemotherapeutic drug in addition to radiotherapy and supportive treatments. Therefore, there is a need to develop more effective new treatment strategies in metastatic prostate cancer patients. In this study, it was aimed to investigate the cytotoxic and apoptotic effect of ruteakarpine, which is known to have an anticarcinogenic effect by inhibiting both COX-2 and topoisomerases, and to investigate its potential therapeutic effect. MATERIAL AND METHOD: The therapeutic effect of rutecarpine on the PC-3 human metastatic prostate cancer cell line was evaluated by viability (WST-1), apoptosis (annexin V), cell cycle, AO staining and caspas-3 (ELISA) assays. RESULTS: When compared to the control group, the viability percentages of different concentrations of rutecarpine (5, 10, 20 and 40 μM) after 24 and 48 hours treatment of PC-3 cells, it was determined that administration of 20 and 40 μM rutaecarpine for 48 hours most effectively decreased cell viability (p <0.05) . After 48 hours of treatment, it was observed that early and late apoptosis rate increased significantly compared to the control group due to the increasing dose by both Annexin V analysis and AO staining (p <0.05). In addition, the amount of active caspase-3 increased significantly in PC-3 cells due to the increasing concentration of ruteakarpine by ELISA analysis (p <0.05). CONCLUSION: In this study, it has been shown that ruteakarpine has a cytotoxic and apoptotic effect on PC-3 cells and may be a potential therapeutic agent. In case an anti-tumor effect is detected by in vivo studies; We believe that the use of ruteakarpine alone or in combination with routine chemotherapeutic agents may support the treatment.
