Abstract (EN)

The skin is a barrier organ that has various physiological functions as well as protecting the body against harmful external factors. Besides, the primary condition of human desire to be beautiful, well-groomed and attractive is to have a healthy skin. Changes in skin health that lead to impaired skin integrity are called wounds. Wound healing consists of three consecutive, highly complex stages. Firstly, inflamation stage with immediate responses such as prevention of blood loss and microbial contamination, occours immediately and lasts approximately 48 hours. Then, the new tissue formation phase, which is the fundamental of wound healing, begins. Finally, the randomly formed proteinic structures are restored in remodelling phase. Elimination of factors that interfere with wound healing mechanisms, such as oxidative stress, will increase the rate of wound healing and the possibility of perfect recovery of the skin to its original state. It is known that rutin will have a positive effect on healing, especially in the first and second stages of the wound healing process, due to its free radical scavenging and collagen production-supporting properties. However, various molecular disadvantages such as low solubility reduce the therapeutic potential of Rutin hydrate. Thanks to its increased surface area, nanofibers help to eliminate the low solubility problem and show the necessary skeletal structure for cell progression, which helps regeneration thanks to its porous structure. Within the scope of this thesis, Rutin-hydrate loaded nanofibers were developed by electrospinning method and evaluated in vitro and in vivo for use as a wound dressing.
