Determination of the antibacterial effect of acidiphilini from the reproductive system of healthy women on bacteria causing urinary tract infection

Abstract (EN)

The current study was aimed to estimate the activity of Lactobacillus supernatant against pathogenic bacteria that isolated from urinary tract infection in women. Swabs and samples were taken from clinical sources, including samples of vagina and urine at June to October 2023. With regard to clinical samples, names, ages and health status were taken for volunteers in the study. Lactobacillus acidophilus isolate was obtained from 50 urogenital tracts of healthy woman. Vaginal swab was cultured on chocolate agar, blood agar, and MacConkys agar (as routine work), and incubated at 37◦C for 24-48 hours under 5% carbon dioxide conditions. While, Uropathogenic bacteria isolates were obtained from 178 females (23-45 years) Inpatients in the Maternity and Children's Hospital, with urinary tract infections, since June to October 2023, urine samples obtained by med stream urine specimens technique. Urine samples were examined microscopically and cultured on chocolate agar, blood agar, and MacConkys agar by loop inoculation for quantitation of organisms in urine, the plates were incubated at 37◦c for 24-48 hours. The current study included 178 samples including. The findings were found that 102(57.3%) of total samples were appear as positive results for bacterial growth that cultured optimal cultured media such as blood agar, mantol agar, and MacConkey agar. 76(42.7%) of total samples were appear as negative results for bacterial growth. The results show the numbers and percentages of isolates for each type of bacteria that were isolated from clinical and environmental sources, where it is noted that E. coli was the most isolated, with 31(30.4%) isolates out of a total of 102 isolates, followed by S. aureus as the number of its isolates reached 19(18.6%). While, Enterobacter cloacae the least isolated, as the number of its isolates reached 11(8.8%) out of a total of 102 isolates. For antibiotic susceptibility test, E. coli showed total resistance (100%) to Amoxicillin/clavulanic acid and Ampicillin. 40% resistance to Nalidixic. Otherwise, E. coli showed 30%, 10%, 50%, 20% and 60% resistance to Imipenem, Amikacin, Gentamicin, Azithromycin and Cefepime respectively. Klebsiella spp. showed total resistance (100%) to Amoxicillin/clavulanic acid and Ampicillin. 40% resistance to Nalidixic. Otherwise, Klebsiella spp. showed 10%, 0%, 10%, 40% and 20% resistance to Imipenem, Amikacin, Gentamicin, Azithromycin and Cefepime respectively. P. aeruginosa showed total resistance (100%) to Amoxicillin/clavulanic acid and Ampicillin. 30% resistance to Nalidixic. Otherwise, P. aeruginosa showed 70%, 70%, 70%, 20% and 70% resistance to Imipenem, Amikacin, Gentamicin, Azithromycin and Cefepime respectively. E. faecalis showed total resistance (100%) to Amoxicillin/clavulanic acid and Ampicillin. 50% resistance to Nalidixic. Otherwise, E. faecalis showed 20%, 10%, 30%, 30% and 40% resistance to Imipenem, Amikacin, Gentamicin, Azithromycin and Cefepime respectively. Staphylococcus spp. showed total resistance to Penicillin, Oxacillin and Cefoxitin. 20%, 0%, 20%, 40%, 10%, 80% and 40% resistance to Gentamicin, Nitrofurantoin, Erythromycin, Azithromycin, Refampicin, Trimethoprim and Ciprofloxacin respectively. Finally, the zone of inhibition of Lactobacillus supernatant against the study bacteria, where the results show that E. faecalis was the most sensitive towards the Lactobacillus supernatant, as the inhibition zone reached 41.2 mm, while S. aureus was the least sensitive towards the Lactobacillus supernatant, as the inhibition zone reached 19.8 mm.
