2. ABSTRACT The aim of this study was to investigate the effects of paxillin and iberiotoxin on calcium-activated potassium channels in cultured rat dorsal root ganglion neurones. Dorsal root ganglia were dissected out and isolated as single cells by enzymatic and physical procedures and placed in humidified incubator containing % 5 CO2 at 37° C. Actions potentials were recorded from these cells using the whole cell current clamp mode of the patch clamp technique. Reversial potential of the calcium- activated potassium channels were determined. Application iberiotoxin and paxillin significantly inhibited afterhyperpolarisation in a dose dependent manner. Result from this study indicates that, iberiotoxin and paxillin inhibits action potential after hyperpolarisations in cultured rat dorsal root ganglia neurones by blocking calcium- activated potassium channels.
