Invitro investigation of relationships between leptin and carbonic anhydrase III in rat adipase tissue

In vitro Investigation of Relationships Between Leptin and Carbonic Anhydrase III in Rat Adipose Tissue. Leptin, a peptide hormone with endocrine, paracrine and autocrine characteristics, is synthesized by adipose tissue, It has antilipogenetic properties and decreases intracellular lipid content by regulating the expression of some enzymes. Carbonic anhydrase III is the most abundant intracellular protein of rat adipose tissue. Its function in this tissue is unclear, but, it has been shown that its amount decreased together with obesity and this decrease might be caused by insulin. CA V has been reported to have a role in insulin secretion after an in vitro study on rat pancreas cell culture. In this research it was intended to show the effects of leptin on expression of CA III and of CA inhibitor dorzolamide on leptin secretion in rat fat tissue explants and cultured cells. Since, the influence of insulin and dexamethasone on leptin secretion from rat fat tissues has been investigated by many researchers previously, these hormones was included just for the confirmation of the procedures involved, and, also for their roles in energy metabolism. The experiments made has shown that insulin and dexamethasone increased leptin secretion and CA activity, and CA inhibition caused a decrease in leptin secretion. According to the results of fat cell culture experiments, it was found that CA expression has been decreased by the action of leptin, increased by insulin and caused no significant change by dexamethasone. It was concluded that CA III had a role in the mechanism of leptin secretion and that the decrease in CA III observed in obesity has been caused by leptin.
