Effects of sodium selenate and vitamin E in inflammation of rat prostate and vas deferens tissue obtained with bacterial lipopolysaccharide

ABSTRACT Effects of sodium selenate and vitamin E in iflammation of rat prostate and vas deferens tissue obtained with bacterial lipopolysaccharide In the last ten years there are experimental and clinical investigations on antioxidant substances. These ivestigations show that, in time they will take place in treatment protocols. The restorative effects of vitamin E and sodium selenate, on inflammation of prostate and vas deferens have been aimed to ivestigate. Two main groups (control and LPS given) of Wistar Kyato rats, with a mean weight of 246gr were used. Inflammation was induced in LPS given group (n=15), with intraperitoneal injection of LPS (10mg/kg). LPS given group was subdivided in three groups. In the first subgroup (n=6) single dose of LPS and aminoguanidin (100mg/kg), (ip); in the second subgroup (n=6) three days of single dose of intramuscular (im) vitamin E (0.3mg/kg/day) and in the fourth day LPS and vitamin E together, in the third subgroup (n=6) three days of single dose sodium selenate (16ug/kg/day) (ip) and in the fourth day LPS and sodium selenate were administered together. In the control group (n=15) isotonic solution (O.lcc/lOOgr) (ip) were administered. There were four control subgroups. In the first control subgroup (n=6), single dose of aminoguanidin; in the second group (n=6) four days of vitamin E and in the other subgroup four days of sodium selenate were given. Vas deferens of all the groups were divided into two 1 cm pieces in the middle as prostatic and epididymal parts. Electrical field stimulation (EFS) (2, 4, 8, 16, 32 Hz, 50 V, 0.5ms) was administered. Difference in contractile responses of vas deferens tissue was defined according to the maximum response obtained with 40mM KC1. These responses were significantly decreased in bacterial lipopolysaccaride-treated rats (P<0.05). However, the differences in selenium and vitamin E treated LPS groups were not significant. There was complete response to antioxidant treatment. The sections from contralateral vas deferens and prostatic tissue were stained with haematoxylin and eosin and inflammatory cells were evaluated microscopically. Inflammation was seen in the prostate and vas deferens in the LPS given group. There was a response in the inflammation to vitamin E in prostate and prostatic part of the vas deferens but there was response to both of the antioxidants in the epididymal part of the vas deferens. These results were confirmed microscopically. In conclusion, a possible increased oxidative activity in the vas deferens tissue may be a major reason for the impairment of contractu responses and inflammation of the tissue. The restorative effects of selenium and vitamin E on this hypocontractilty and microscopic inflammation may depend on its antioxidant properties and partly its inhibitory action on inducible nitric oxide synthase production. In future; we may use antioxidants for treating infertility or prostatitis. There is a need for more investigations about antioxidant therapy in urological disorders.
