Effect of apelin hormone on renal ischemia/reperfusion induced oxidative damage in rats

Abstract (EN)
Apelin is a peptide structure hormone, which is defined as the ligand from APJ receptor. Apelin and APJ, which are known that synthesized in several peripheral tissues and draw attention to the detection of kidney tissue in mammals. This study was carried to investigate three different doses of Apelin-13 effect on the renal ischemia reperfusion (I/R). In the study, 35 Sprague-Dawley male rats were used in total and 5 groups are created (n=7). In Sham (control) group, right kidneys of the animals were dissected. In I/R group, while dissecting right kidneys, 45 min. ischemia and after that 3 h. reperfusion were applied. In the groups in which apelin-13 is applied (1, 10 and 100 μg), different from I/R group apelin-13 was given as intraperitoneal (i.p.) at the outset of the ischemia. BUN, as a sample of the serum, creatinine, sodium, potassium, chloride, total protein, albumin, AST, ALT and GGT autoanalyzer and TNF-α, IL-1β, IL-6, TAS and TOS parameters were specified by ELISA analysis. MDA, SOD, CAT and GSH-Px levels of renal tissue are measured. According to serum biochemical parameters, a significant increment was observed in all groups other than chloride and GGT groups with respect to sham groups. Otherhand, a significant increment was observed in all groups except chloride and GGT group and also that applies to I/R group as well with respect to sham group (p<0.05). Sodium, total protein and albumin parameters decrease significantly (p<0.05). When analyzing apelin applied groups, it is observed data ratios vary according to the dosages in comparison with I/R group. When estimating tissue samples in the context of antioxidant enzymes, with respect to sham group, in I/R group SOD, CAT and GSH-Px decrease dramatically however MDA level increase significantly (p<0.05). In ELISA studies, all parameters increase dramatically except TAS and that applies I/R group with respect to sham group as well (p<0.05). In TAS, a statistically significant decrement was observed. When analyzing apelin applied groups rather than I/R group, a significance was determined especially the groups that 100 μg was applied, (p<0.05). Our results show that implementing apelin-13 in short times partly reduces ischemia damage.
