The healing effects of ganoderma lucidum on mesenteric ischemia-reperfusion injury in rats

Abstract (EN)

SUMMARY Background: İntestinal ischemia reperfusion injury serious clinic problem and carries high morbidity and mortality risk. A various clinic events such as intestinal obstruction, severe trauma and aortic by-pass procedures may cause intestinal ischemia reperfusioninjury. There is no reliable parameter for early diagnosing and single ajan for effective treatment. Ganoderma lucidum is known as immunmodulator, antioxidant and anti inflamatuar. The purpose of this study was to investigate the healing effects of ganoderma lucidum on intestinal ischemia reperfusion injury in an experimental study. Materials and Methods: The study was performed by using 24 male Wistar–Albino rats (weighing 250–300 g) randomly divided into three groups: Group I, sham group; Group II, I/R group; Group III, I/R with previous ganoderma lucidum treatment group. After laparotomy, ischemia and reperfusion were performed for 30 and 90 min, respectively, on all the groups except for sham group. Intestinal and hepatic tissue examples were stained by using routine histological methods and examined under the light microscope. Serum and intestinal and hepatic tissue Malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH‑Px) levels were measured. Result were compared statistically by using SPSS Windows 18 programme. P values <0.05 were accepted as significant. Result: The I/R group had significantly elevated leukocyte, thrombocyte, serum MDA, intestinal tissue MDA and hepatic tissue MDA levels compared with the sham group. The I/R group had significantly reduced serum SOD, Serum GSH-Px, intestinal tissue SOD, hepatic tissue SOD and hepatic tissue GSH-Px MDA levels compared with the sham group. Previously administration of ganoderma lucidum efficiently improved these negative alterations.Ischemia reperfusion injury resulted in significantchanges in intestinal tissue such as impairmentin villus patternand hepatic tissuesuch as sinusoidal dilatation and congestion of I/R group rats. Conclusions: Ourexperimental study showed that intestinal ischemia reperfusioninjury leads to significant oxidative stress by inducing free oxygen radicals in intestinal and hepatic tissues. Serum and tissue MDA, SOD and GSH-Px levels were considerably helpful labaratory parametersto identify oxidative stress. The healing effects of Ganoderma lucidum on intestinal ischemia reperfusion injury was promising in the light of these parameters.
