The effect of mesenchymal stem cells to laminin, calsitonin, AVβ3 integrin in the implantation area of rats

Implantation occurs with the attachment of the embryo to uterus. During the implantation, of structural and morphologic events are occurs in uterus. Laminin, a component of connective tissue, is known to play a role in embryonic development and placenta formation. It is known that the hormone of calcitonin secreted from the thyroid gland has autocrine and paracrine effect on the uterus during implantation. The best characterized cell adhesion molecule in the luminal epithelium of the endometrium is αVβ3 integrin. From this data, we aimed to demonstrate the effect of mesenchymal stem cells to laminin, calcitonin and αVβ3 integrin in the implantation region of rats by immunohistochemical method. In our study 16 adult "Wistar Albino" female rats were divided into two different subgroups as control and experiment groups on the 10th day of gestation. The MSCs which were obtained from rat bone marrow were given to 8 rats. Phosphate Buffered Saline (PBS) was given to 8 rats which were in the control group. The same procedures were performed for 21-day-old pregnant rats. A total of 32 "Wistar Albino" female rats were included to the study and their implantation areas and the placentas were taken. Then, these samples were immunohistochemically assayed for laminin, calcitonin and αvβ3 integrin. It is seen that, there is no significant differences in 10 days pregnant rats for Laminin immunolocalization between control and experimental groups. When the calcitonin was examined for immunolocalization, weak immunoreactivity was seen around the decidual cells in the experimental group but moderate-intensity immunoreactivity in the control group. In the immunolocalization of αVβ3 integrin, moderate-intensity immunoreactivity was observed around the vessel wall and gland epithelium in the control group while weak intensity immunoreactivity was seen in the experimental group. In 21 day pregnant rats; weak immunoreactivity was seen for lamin immunolocalization in the gland epithelium of experimental group while control group showed modarete immunoreactivity in the gland epithelium. There was no significant difference no between the control and experimental groups for calcitonin and αVβ3 integrin immunocalization. As a result; In 21-day-old pregnant rats, laminin immunolocalization showed weak immunoreactivity in the glandular epithelium of experimental group but moderate immunoreactivity was seen in 10-day-old pregnant rats. Immunocalization of calcitonin in stromal cells of 21-day-old pregnant rats showed moderate immunoreactivity but weak immunoreactivity in 10-day-old pregnant rats. In 21-day-old pregnant rats, giant trophoblastic cells showed weak immunoreactivity for αVβ3 integrin immunolocalization. On the 10th day of gestation, giant trophoblastic cells showed moderate-intensity of immunoreactivity for αVβ3 integrin immunolocalization.
