Comparison of the effectiveness of ascorbic acid and magnesium in renal ischemia reperfusion model in rats

Abstract (EN)

Background: Kidney is one of the most affected organs of ischemia reperfusion injury during surgical procedures. The aim of this study is to determine effectiveness of ascorbic acid and magnesium administration seperately or combined before ischemia on the rat model of renal ischemia and reperfusion damage. Materials and Methods: After the approval of the DEU Local Ethics Board of Animal Experiments, 35 Wistar albino male rats ranging in weight from 250-300 g were divided into 5 groups. The Sham Group (n=7) was a laparotomy, 45 min ischemia and 240 min reperfusions were applied to other groups. No drugs were applied to the IR Group (n=7). 250 mg/kg ascorbic acid 1 hour before ischemia in the Ascorbic Acid Group (n=7), Magnesium Group (n=7) was administered to 200 mg/kg magnesium sulfate 1 hour before ischemia, ascorbic acid and magnesium sulfate intraperitoneal in the same doses 1 hour before ischemia (n=7). MDA and GSH levels were determined in the renal tissues received, serum BUN and creatine levels were measured in blood samples. H&E and PAS paintings for histological examinations, TUNEL immunohistochemical staining for apoptotic cell examinations. p<0.05 value was considered statistically significant. Results: The BUN value was found significantly lower in the Ascorbic Acid + Magnesium group compared to the IR group (p:0.021). The value of creatine was found significantly lower in Magnesium group compared to the IR group (p:0,002). The MDA value was found significantly lower in the Group of Askorbit Acid (p:0.034) and Group of Askorbic Acid + Magnesium (p:0.001) compared to the IR group. GSH value of Group of Magnesium (p<0.001) and Group of Ascorbic Acid + Magnesium (p<0,001) were significantly higher in other groups. Less damage was detected in the groups applied to the drug as a result of H&E and PAS paintings, but together it was observed that the application did not constitute an advantage over the individual application. TUNEL counts of the Group of Ascorbic Acid + Magnesium was found significantly lower than the IR group counts (p:0,005). Conclusions: According to the data obtained, it was concluded that the separate and combined of 250 mg/kg ascorbic acid and 200 mg/kg magnesium sulfate applied before ischemia in the rat renal IR model reduces the damage of the kidney.
