Morphologic and molecular characterization of some selected common bean (Phaseolus vulgaris L.) lines and cultivars

This study was conducted to reveal the morphological and molecular characterization of some common bean cultivars and lines collected by Karadeniz Agricultural Research Institute in Terme, Ladik, Tekkeköy and Çarşamba provinces. All lines were grown under Antalya ecological conditions and morphological characteristics of all 36 lines were evaluated in accordance with UPOV criteria while Sequence Characterized Amplified Region (SCAR) and Simple Sequence Repeat (SSR) markers were employed for molecular characterization. Principles Component Analyze of the data obtained from morphological characterization were conducted. Analyze results revealed that first three components can be used to explain the 50 % of the variation among lines and cultivars and this result indicate that genetic variation among bean lines and cultivars is not high. In terms of molecular characterization while 27 % of SSR primer pairs were monomorphic while 73 % of them were polymorphic. Polymorphism information content (PIC) of SSR primer pairs varied between 0.047 and 0.373. On the other hand all SCAR primers used in this present study were found to be polymorphic and PIC values of SCAR primers were between 0.071 and 0.379. According to statistical analyses results genetic similarity index among bean cultivars and lines showed variation between 0.52- 0.98. Results revealed that there were two main groups among the bean lines and cultivars. In the first group was consisted of Cornell and Widusa cultivars, which belong to Mesoamerican gene pool and in disease resistance differential set. Kabon cultivar, belong to Andean gene pool member and in anthracnose differential set , and Redland Pioneer cultivar belong to Andean gene pool, took a place with other all cultivars and lines used in the present study. This result may indicate that all selected lines may have on close genetic relationship with Andean gene pool. Genetic similarity index of the second group varied between 0.71 and 0.98. In a comparison between dendrogram constructed by using morphologic and molecular analyses data it is possible to say that molecular data much more reliable than morphologic data.
