Abstract (EN)
Non-syndromic cleft lip/palate (CL/P) malformation is one of the common congenital defects. Environmental and genetic factors in the formation of the disease is avaliable in the literature. Previous studies of mutations in the MSX1 gene non-syndromic cleft lip and/or palate has contributed to the formation. In this master?s thesis, the candidate gene MSX1 gene G267C region, were analyzed for its association with the non-syndromic cleft lip and/or palate malformation. The blood samples used in this study, admitted to, Ankara University, Faculty of Dentistry non-syndromic cleft lip and/or palate were obtainned from Turkish patients. Peripheral blood samples from patients, phenol chloroform method was applied for genomic DNA isolation and PCR metod was enforced. After then, RFLP metod applied to the resulting products, mutations determined, gene products was cut with restiriction enzymes. Enzyme products were photographed by performing agaroz gel electroforesis. As a result, 4 heterozygous region were determined in G267C region of the MSX1 gene. This findings, support the role of G267C convention in the MSX1 gene to non-syndromic cleft lip and/or palate in Turkish population.
