The comparative investigation of cytotoxic and genotoxic properties of synthetic commercial tissue adhesives

Abstract (EN)
Today, bioadhesives are widely used. There is no detailed study of cytotoxic and especially genotoxic properties of synthetic tissue adhesives based on different chemical bases widely used in daily life and in surgery. Therefore, investigations of their safety, including cytotoxic and genotoxic potentials, have a vital prospect for revealing the harmful effects after their use. Within the scope of this doctoral thesis, the cytotoxic and genotoxic properties of CMT, DYC, LEU, which are commonly used synthetic-based commercial tissue adhesives based on 4 different chemical bases, as well as PU-A, which we synthesized in our laboratory, were examined in vitro. The modulus values of CMT, DYC, LEU, and PU-A tissue adhesives were found 1.659±0.594, 2.580±1.709, 2.402±0.720, and 2.489±1.246 kN/mm², respectively. According to the cytotoxicity test results on L-929 cells, CMT, DYC, LEU, and PU-A tissue adhesives compared to the control group were 96.05±9.89, 57.2±5.69, 95.11±4.95, and 100.34±4.47 for 24 hours, and they also have 80.4±3.75, 0±0, 69.66±11.39, and 63.25±11.5 percent cell viability values for 48 hours, respectively. For the cytotoxicity test results on HepG2 cells, CMT, DYC, LEU, and PU-A tissue adhesives compared to the control group were 72.66±9.67, 56.42±6.36, 103.15±11.14, and 95.61±11.74 for 24 hours, and they also have 81.02±11.19, 0±0, 66.57±8.69, and 70.53±5.58 percent cell viability values for 48 hours, respectively. In the comet assay performed as a genotoxicity test, the values of (-) Control, (+) Control, CMT, DYC, LEU and PU-A groups on L-929 cells in terms of Tail DNA (%) were 6.97, 23.52, 30.5, 22.95, 19.06, and 29.18, respectively. On the other hand, the values of (-) Control, (+) Control, CMT, DYC, LEU and PU-A groups on HepG2 cells in terms of Tail DNA (%) were 11.72, 31.88, 20.89, 28.47, 38.83, and 37.76, respectively.
