Abstract (EN)

Ischemia-reperfusion (IR) injury is a series of pathophysiological tracks with recirculation instead of oxygen. It is known that the brain can adapt to unexpected events at a subletal dose and improve its resistance to its cells. Remote ischemic postconditioning (RePostC) is a noninvasive method that protects vital organs sensitive to ischemia by interventions in remote organs. It was aimed to effects of remote ischemic postconditioning on infarct area, providing protection from apoptosis and necrosis for oxidative stress, energy modulator, Asprosine, Nuclear Factor Erythroid-Related Factor 2 (Nrf2) that mediates neuroprotection by regulating antioxidant genes in cerebral ischemia, miRNA-93 that can reduce inflammatory response and apoptosis and miRNA-27b levels, which is thought to support recovery after ischemic stroke by regulating activated protein kinase activity in cerebral IR injury. In our study, rats were divided into 3 groups. Ischemia for 90 minutes and reperfusion for 24 hours was applied to the middle cerebral artery (OSA). RePostC was applied to the right hind leg as IR for 3 cycles of 5 minutes each. The area of infarction was determined by Tetrazolium Chloride staining. Nrf2 protein expression levels were measured by Western Blot, miRNA levels by PCR and Asprosin levels by ELISA method. While the infarct area, miRNA-93 and miRNA-27b levels increased in IR, they decreased significantly with treatment. Asprosin and Nrf2 levels decreased in IR, but increased significantly with RePostC. Asprosin, Nrf2, miRNA-93 and miRNA-27b levels may be biomarkers in the diagnosis of IR injury and evaluation of treatment. RePostC; By affecting asprosine and Nrf2 levels, it can reduce IR-induced oxidative stress, inflammation, apoptosis and necrosis. These effects may be mediated by miRNA-93 and miRNA-27b. RePostC, which is a noninvasive, safe and easily applicable method, can be evaluated as a treatment approach in cerebral IR injury and can be clinically tested.
