SUMMARYResearch of Chlamydia trachomatis Infections With Polymerase ChainReaction and Direct Fluoresan Antibody Method in servical samples(Dr. Alper KAPDAGLI)Chlamydia infections are among most common bacterial infections andcost for worldeconomy. Many countries developed national survey studies for riskgroups to avoideconomic cost. Women at fertile age with mucopurelent vaginal dischargeand sex workersare in risk groups. The aim of our work was to determine the prevalenceof Chlamydiatrachomatis infections among risk groups in Aydın area by a surveystudy and to establish abase for further studies.Forty-nine sex workers from Aydın brothel and 47 patients attended ADUGynecologyand Obstetric Department and Gynecology and Obstetric Hospital withmucopurelentdischarge, and 44 patients without mucopurelent discharge as control, atotal of 91 15-45years-old patients were included to this study. Endocervical samplesfrom the study groupswere studied by PCR and DFA for presence of C. trachomatis. Cervicalsamples from thesame patients were cultured on blood agar, Eozin metylene Blue (EMB)agar, chocolate agarand Sabouraud dextrose agar (SDA), Gram staining and direct microscopicanalysis were alsodone. Two samples were taken with swabs from posterior fornix and onewas used for pHmeasurement and the other one for direct microscopy and Whiff test.One sample from patient was positive by PCR and the prevalence wascalculated as%0.7 (n=140). The prevalence for the study group was calculated as(n=140) % 0.7. The DFAtest was negative for all patients tested.Culture results; of 140 samples 100 (%71.4) had normal vagina flora,and 19 (%13.5)had no growth, 8 (%5.7) had Gardnerella vaginalis, 7 (%5) had Candidaalbicans, 3 (%2.1)had Streptococcus agalactiae, 3 had (%2.1) Escherichia coli, and one(%0.7) had Neisseriaegonorrhoeae. Fifteen (%10.7) patients were diagnosed as bacterialvaginosis. Trichomonasvaginalis trofozoits were observed in samples from 3 (%2.1) patients bydirect microscopicanalysis.In our study based on PCR endocervical sample from one person found tobe positiveand the prevalence was 0.7% (n=140). DFA test were negative for all 127samples with a cutoff value of >10 elementary body in each field. The prevalence wasincreased to 7.1% and to16% when the cut-off is accepted as 6-10 EB and 2-5 EB in each fieldreach, respectively.In Aydın area the prevalence of C. trachomatis among sex workers andwomen withmucopurelent discharge change in relation with the method and the cutoff value used. Furtherand larger survey studies should be done to obtain safe data.
