Wound healing effects of sweetgum oil in cell culture

Abstract (EN)

Considering the researches and practices for wound treatment all over the world, it is clear how valuable a successful wound treatment is. Today, the discovery of new effective agents increases the quality of life of patients by shortening the healing process. The wound healing effects of Sığla oil (Sweetgum oil), which is the product of Liquidambar orientalis grown in our country, were tested in cell culture medium and its mechanism of action was investigated in the study. In vitro experiments were applied to human derived HaCaT keratinocyte cells lines which cultured under appropriate conditions. Sweetgum oil belonging to two different local producers was brought from Muğla. Centella asiatica plant extract, which has proven wound healing effects, was used as a positive control for the comparison of the efficacy of sweetgum oil. Methods which applied to determine the effects of the extract on wound healing is, firstly MTT for non-toxic concentration determination for evaluation of the efficacy of sweetgum oil in HaCaT keratinocyte cells; real time cell analysis system and scratch test for proliferation and cell migration detection; and real time polymerase chain reaction to check COL1A1 (collajen type-1 alpha-1) mRNA expression. In the study, non-toxic concentrations were determined as 0,1 μg/ml (100 ng/ml), 0,01 μg/ml (10 ng/ml) and 0,001 μg/ml (1 ng/ml) as a result of MTT method. According to the real time cell analysis system results, a significant increase in cell proliferation and migration was observed at 0,001 μg/ml concentrations in a time (72 h) dependent manner. According to the scratch analysis, at the end of the 48th hour it was observed that the distance on both sides of the wound was significantly closed at 0,001 μg/ml concentrations in a time-dependent manner. According to the Real-Time PCR result, no increase in COL1A1 (collajen type-1 alpha-1) mRNA expression levels was observed. In the study, it was concluded that sweetgum oil is effective in wound healing in vitro.
