43 8- SUMMARY Study of Bovine Herpes Virus 1 (BHV-1) Proteins with Immunoperoxidase, Electrophoresis and Western Immunoblot Methods. Bovine herpes virus 1 (BHV-1) proteins were studied with three different methods. Initially, Immunoperoxidase (IP) test was used to investigate the intracellular localization of viral proteins at different stages of BHV-1 infection. Viral proteins were determined to be in the nucleus of infected cells at the early hours of infection. It was noted that, as the infection progressed, viral proteins started to accumulate in the cytoplasm. In order to analyse BHV-1 proteins by SDS-PAGE, BHV-1 infected MDBK and control MDBK cell lysates were subjected to electrophoresis on 10 % polyacrylamide gel. After staining with Coomassie blue, 50 different proteins were determined in the infected MDBK cells, whereas only 20 different proteins were observed in the control MDBK cells. However, precise analysis of viral proteins could not be carried out. Infected MDBK and VERO cell proteins, subjected to electrophoresis, were also examined by Western blot using immune rabbit serum. As a result, proteins at the molecular weights of approximately 55, 75 and 180 kDa were determined to be immunogenic proteins of BHV-1.
